Transformation of Streptomycin Resistance in Mycobacterium phlei by DNA from Streptomycin-resistance Strain


Abstract
Experimental studies on the transformation of streptomycin (S`.4) resistance in Jlycobacterium (M) phiei were carried cut with DNAR derived from S=Vi-resistant culture.
Two methods were applied for t¢¥-.e study; one was culture method in which the bacterial cells to be trans= ormed were culture/ with DNAR, and the other was resting cell method in which the cells were treated with the DN AR at C C and 37C.
After the treatment, cells were cultured on media containing Si~1 in serial concentrations to determine the increase in the resistance. The results were as follows.
1. The SM resistance of DNAR-treated cells ; by culture method was markedly increased up to 1, 000 - 5, G00 mcg/ml as compared with the original cells (i,, mcg; :nl) .
2. No significant SI~I resistance was observed by the resting cell method treated at 0C.
3. The :esistance to Sli increased up to 5, C~0 mcg/r_21 in. cells washed after treatment with DNAR at 37C ands subcultured en original media. I¢¥_owever, unwashed cells did not show any significant increase in the resistance.
4. It was conceivable with the results obtained that SM resistance can be transformed in . M. phlei.
